Azapin Indications
Schizophrenia. Treatment of patients with schizophrenia who have no effect on the application of "classical" antipsychotics or celebrated their intolerance.
Lack of effect is defined as a lack of satisfactory clinical improvement despite treatment with at least two antipsychotics, which are used in adequate doses for an appropriate period.
Intolerance is defined as the inability to achieve adequate clinical improvement using the "classic" antipsychotic drugs due to severe adverse reactions, which are not amenable to correction (extrapyramidal side effects or tardive dyskinesia).
The risk of recurrent suicide attempts. To reduce the risk of recurrent suicidal behavior in patients with schizophrenia or schizoid disorders are prone to chronic risk of repeat suicide attempts, known in the history of the current clinical status of the patient.
Psychosis during therapy of Parkinson's disease. Mental disorders are observed during the treatment of Parkinson's disease, in cases where the standard therapy was ineffective.
Co0ntradictions
Hypersensitivity to clozapine or any other components of the formulation; toxic or idiosyncratic granulocytopenia or agranulocytosis in history (with the exception of granulocytopenia or agranulocytosis as a result of previously used chemotherapy); inability to regularly monitor the condition of the patient's blood; impaired bone marrow function
Dosage & Administration
Dose is chosen individually. Each patient should use the minimum effective dose. In patients receiving drugs that interact with Azapinom (such as benzodiazepines and selective serotonin reuptake inhibitors), it is necessary to make changes in the dosing.
For oral dosage recommended for the following:
Schizophrenia, treatment-resistant
The initial phase of treatment. At day 1 clozapine 12.5 mg administered 1 or 2 times per day; the 2nd day - 25 or 50 mg of clozapine. Further, provided good tolerability, the dose can slowly increase of 25-50 mg, so that for 2-3 weeks to achieve a daily dose of 300 mg. Then, if necessary, the daily dose can be increased further by 50-100 mg every 3-4 days or better - every 7 days. In elderly patients it is recommended to begin treatment with a particularly low dose (1 st day - 12.5 mg once daily), and the next increase limited dose of 25 mg / day.
The therapeutic dose range. In most patients, the onset of antipsychotic drug action can be expected when using a daily dose of 300-450 mg Azapina (in divided doses). Some patients may also be effective lower dose, others may require doses up to 600 mg / day. The daily dose can be distributed unevenly in the individual techniques, assigning it a large part of the bedtime.
